AstraZeneca and Daiichi Sankyo have entered into a clinical collaboration agreement with Summit Therapeutics to evaluate Datroway (datopotamab deruxtecan) in combination with ivonescimab across multiple tumour types, including lung and breast cancers. The clinical programme is expected to begin with a Phase III trial in first-line triple-negative breast cancer.
Datroway is a specifically engineered TROP2-directed DXd antibody-drug conjugate discovered by Daiichi Sankyo and jointly developed and commercialised with AstraZeneca. Ivonescimab is a bispecific antibody designed to combine PD-1 blockade with VEGF inhibition in a single molecule.
Under the agreement, each company will supply its respective medicine for the planned combination trials, which will be sponsored by AstraZeneca or Daiichi Sankyo. The three companies will contribute to trial costs and retain the development and commercial rights to their respective products.
The collaboration forms part of a broader development strategy focused on combinations of antibody-drug conjugates and next-generation immunotherapies. It follows a separate agreement between AstraZeneca and Summit Therapeutics to investigate ivonescimab with sonesitatug vedotin in gastrointestinal cancers, as well as an equity investment by AstraZeneca intended in part to support clinical collaborations involving ivonescimab.
Ivonescimab is approved in China for certain patients with non-small cell lung cancer. A Biologics License Application for EGFR-mutated non-small cell lung cancer is also under review by the US Food and Drug Administration. The medicine is being investigated in Phase III studies across several solid tumours, including lung, breast, biliary tract, bladder, colorectal, pancreatic, and head and neck cancers.
